Understanding of heart machinery and metabolic responses of cod and halibut following oil compounds exposure

1 Purpose
The objective of this study is to assess the effect of oil compounds (e.g. phenanthrene and its alkylated paralogs) on the metabolic activity of two fish species, Atlantic cod (gadus morhua) and Atlantic halibut (Hippoglossus hippoglossus). Some oil compounds have been shown to have acute but reversible effect on cardiac function and therefore oxygen consumption (metabolism).

2 Distress
Moderate stress will be performed to the fish by manually chasing them to force the fish to swim and reach a higher metabolic rate. Fish will be then placed and resting in respirometry chambers to measure its oxygen consumption. A group of fish will be also exposed to oil compounds in order to mimic an environmental stress condition and assess the impact on their metabolic activity.

3 Expected benefit
This study will increase knowledge about fish stress and metabolic physiology and how the fish will handle/adapt to a potential oil discharge into environment.

4 Number of animals, and what kind
60 Atlantic cod and 60 Atlantic halibut.

5 How to adhere to 3R
All measurement will be undertaken in living animal placed in their respective rearing tank. Limited number of fish will be used for experiment. The number of fish is used to get a valuable statistical
analysis.
We will terminate the experiment if the fish experience humane endpoints (see description). If the fish reach humane endpoints, euthanasia will be performed with overdose of MS222 and the experiment will be stopped.
