Role of AQP4 in health and disease: Non channel functions of AQP4

Our laboratory has its focus on the role of aquaporin water channels. Aquaporin-4 (AQP4) is the predominant brain water channel. We have generated a transgenic mouse strain lacking AQP4 (AQP4-KO), which we have used in several of our projects.
In this project we will be looking at a newly generated mouse with a single mutation in the AQP4 gene. The mouse is generated by Transgensenteret at IMB, and the mutation is based on a patient with a similar mutation. As we don't know to what extent the mice will have a phenotype, we will be investigating all the organs where AQP4 is expressed, including the central nervous system ( brain, spinal cord), eyeballs, kidneys, heart and skeletal muscle. The mutant mice will be backcrossed with WT C57/Bl6 mice for 7 generations.
To begin with, we will perform investigate the morphology (microscopy, immunohistochemistry), mRNA and protein expression (Western blotting and qPCR), and generation of primary cell cultures.
We will need to build up a breeding colony of Het x Het over time. We will also perform behavioral tests and functional experiments of brain and heart. Details of these experiments will follow in future applications.
