Further characterization of TG2-autoreactive B cells in 14E06 Ig KI mice

The purpose of the experiments described in this application is to further characterize TG2-specific B cells in '14E06' immunoglobulin knock-in mice. This will be crucial to understand the mechanisms behind the formation of autoantibodies in patients with celiac disease.
The distress for the animals will be minimal. Mice are expected to develop mild pain for a short amount of time due to subcutaneous, intraperitoneal and intravenous administration of antigen or cells as well as the withdrawal of blood. The injected antigens will not cause any discomfort to the animals, volumes are kept small and blood samples will be taken minimum 7 days apart.
The number of animals per group are kept at a minimum that is necessary to obtain statistically significant difference. Moreover, pilot experiments aimed at further reducing the number of mice in experiments are included in the application.
